What tesamorelin doses do people report using?
1 mg and 2 mg recur in the first-person reports reviewed here. Some authors describe beginning with 0.5 mg or 1 mg; others report changing the amount after blood tests. Both daily use and five-days-per-week schedules appear. The table identifies the actual account behind each example.
These are selected, unverified personal accounts, not a survey, recommended protocol, or proof that the featured research products are suitable for human use. Amounts refer to tesamorelin; co-use is identified separately.
| Reported amount and frequency | Time reported | Context |
|---|---|---|
| 1 mg daily, then 2 mg daily | Nearly finished 12 weeks | The commenter explicitly reported no days off; no lab results supplied.C1 |
| 0.5 mg daily for 2–3 weeks, then 1 mg daily | About eight weeks of use | Reported IGF-1 and glucose before and during use.C2 |
| 1 mg nightly, then 2 mg nightly, then 0.6 mg with ipamorelin | Seven weeks into a planned 12 | The final combination included 200 mcg ipamorelin; hand numbness was reported.C3 |
| 1 mg daily, fasted morning | Completed about three months | Also used 300 mcg ipamorelin, a calorie deficit, creatine, and substantial training.C4 |
| 2 mg daily, later 1 mg five days weekly | Blood test after three weeks at 2 mg | The commenter changed the schedule after a high IGF-1 result and a break.C5 |
How long do people run tesamorelin cycles?
The accounts include an eight-week update, near-completed or planned 12-week courses, and a completed three-month course. Those are different milestones: the eight-week author was reporting progress, not documenting a finished eight-week cycle.C1C2C4 Another commenter described pausing for about one and a half weeks after a high IGF-1 result, then restarting at 1 mg five days weekly.C5
How much tesamorelin does that add up to?
| Reported schedule pattern | Per week | Eight weeks* | 12 weeks* |
|---|---|---|---|
| 1 mg on five days each week | 5 mg | 40 mg | 60 mg |
| 1 mg every day | 7 mg | 56 mg | 84 mg |
| 2 mg every day | 14 mg | 112 mg | 168 mg |
*Arithmetic assuming the same amount throughout, with no missed days. These are not the exact totals consumed by authors who changed amounts, and they exclude discarded solution.
Five weekly administrations use about 29% less material than seven at the same amount. That explains a cost difference; it does not establish equal benefit. These reports also do not show that non-daily schedules or pauses “reset receptors.” A useful distinction is what someone did versus what comparative research has established.
Do people take tesamorelin in the morning or at night?
Both timings appear in the accounts. The seven-week IGF-1 update described nightly use. The completed three-month combination report used a fasted morning injection.C3C4
These are documented timing choices. They do not provide a head-to-head comparison proving that bedtime produces more GH, or that everyone needs the same fasting window. The morning account also included ipamorelin and major diet and training changes, so its results cannot isolate timing.
Does tesamorelin increase your own HGH production?
Yes. Tesamorelin stimulates the GHRH pathway, prompting the pituitary to release growth hormone. In a small study, 13 healthy men received 2 mg daily for two weeks. Frequent overnight blood sampling showed increased basal and pulsatile GH secretion; IGF-1 increased as well. This demonstrates a biological response, not an ideal long-term bodybuilding dose.C6
What IGF-1 changes did community users report?
| Account | Reported IGF-1 change | Important context |
|---|---|---|
| Eight-week update | 203 → 347 ng/mL | 0.5 mg followed by 1 mg daily; glucose 93 → 100 mg/dL.C2 |
| Seven-week update | 189 → 373 ng/mL | Changed amounts and added ipamorelin; reported hand numbness.C3 |
| Three-week comment | About 174 → 448 | 2 mg daily; the author said the laboratory flagged the result.C5 |
IGF-1 is a downstream response to GH signaling, not a direct HGH reading. These numbers cannot be converted into an equivalent number of HGH injection units. A larger increase is not automatically a better outcome; age-adjusted laboratory ranges and adverse effects matter.C7
How can you tell whether the GH response is useful?
Separate the laboratory response from the result you wanted. An IGF-1 increase does not itself prove better recovery, new muscle, or reduced visceral fat. Compare baseline and follow-up results using the laboratory’s age-adjusted range; glucose and HbA1c also matter. The official label calls for glucose assessment and IGF-1 monitoring.C7
The three-month combination account reported better sleep and visible changes around month two, but also substantial exercise and a calorie deficit.C4 Swelling or hand numbness deserves assessment, rather than being treated as proof that a GH-focused protocol is working. The side-effects guide covers these concerns.
Tesamorelin 10 mg reconstitution: how does the volume change concentration?
Concentration equals the amount of peptide divided by the final solution volume. These examples explain the arithmetic; they do not specify a suitable diluent, sterile preparation, shelf life, or injection regimen for a research vial.
| Peptide amount | Final solution volume | Calculated concentration |
|---|---|---|
| 10 mg | 1 mL | 10 mg/mL |
| 10 mg | 2 mL | 5 mg/mL |
| 10 mg | 3 mL | Approximately 3.33 mg/mL |
Increasing the volume dilutes the solution; it does not create additional peptide. At 5 mg/mL, a 0.1 mL sample contains 0.5 mg. At approximately 3.33 mg/mL, the same 0.1 mL sample contains approximately 0.333 mg. The volume is identical, but the amount is different.
Use the final volume, rather than assuming it always equals the water added. For a labeled medicine, its stated finished concentration takes precedence. WR, for example, specifies 8 mg/mL after its prescribed preparation.1
Tesamorelin dosage calculator: what do the units mean?
On a U-100 syringe, 100 marked units represent 1 mL. Ten units therefore represent 0.1 mL; 16 represent 0.16 mL; 35 represent 0.35 mL. These are volume conversions, not tesamorelin doses. Do not assume every small tick represents one unit: read the actual syringe scale.
For a U-100 scale: volume in mL × 100 = marked units. Convert micrograms first: 1,000 mcg equals 1 mg.
Enter a verified concentration and an amount already specified for the intended preparation. A calculator can check dimensional arithmetic; it cannot choose a treatment dose, confirm vial contents, or establish that a product is suitable for injection.
Can you convert the answer to pen clicks?
Only if the device’s delivered volume per click is known. Milliliters, U-100 markings, and pen clicks are different measurements. The featured pen lists 10 mg in 3 mL, but that concentration alone does not reveal its click calibration.
Where is tesamorelin injected?
The approved instructions use a subcutaneous injection into the abdomen, with the site rotated. The exact product and supplied equipment determine the preparation and injection procedure.5
Rotate the site rather than repeatedly using one spot. Avoid the navel, bruises, scars, irritated or infected skin, and hard bumps left by earlier injections. Use a fresh sterile syringe and needle, clean the skin, and place used sharps in an appropriate disposal container.5
The supplied device instructions matter: a syringe marked in milliliters, a U-100 syringe, and a pen with clicks use different scales. A familiar-looking mark is not enough to establish the amount of medicine.
How do prescription EGRIFTA doses compare?
EGRIFTA WR uses 1.28 mg daily; EGRIFTA SV uses 1.4 mg daily. The older formulation used in the pivotal trials delivered 2 mg daily. Those numbers belong to different formulations, rather than a starting dose followed by two increases.123
| Formulation | Daily amount | Volume per dose | Seven-day total |
|---|---|---|---|
| EGRIFTA WR | 1.28 mg | 0.16 mL | 8.96 mg |
| EGRIFTA SV | 1.4 mg | 0.35 mL | 9.8 mg |
| Original trial formulation | 2 mg | Depends on that formulation | 14 mg |
The weekly totals are arithmetic: daily milligrams × seven. Treatment in these sources is once daily; the total is not a weekly injection.
WR and SV are not interchangeable. Their different doses produce exposure comparable to the original formulation. The volume beside each dose applies only to that product’s labeled preparation.12
What about a 10 mg vial? Ten milligrams describes the container’s total contents. The research vial and pen featured on this site are separate from EGRIFTA. Their packaging does not establish a human dose or validate the instructions above.
The pivotal clinical program assessed visceral-fat outcomes at 26 weeks, with follow-up through 52 weeks. Those study periods differ from the shorter community courses described above.34
How do you reconstitute tesamorelin?
Match the vial name before choosing the liquid or volume. Have a pharmacist or clinician demonstrate the supplied equipment before first use. Work on a clean surface, wash and dry your hands, check expiry dates, and clean the vial stoppers.5
EGRIFTA WR: weekly preparation
11.6 mg vial + 1.3 mL supplied diluent- Use the supplied bacteriostatic water and mixing syringe.
- The instructions draw 1.5 mL, then remove air and excess liquid until exactly 1.3 mL remains.
- Transfer that volume into the vial; discard the mixing syringe safely.
- Swirl gently without shaking. Allow foam to settle; reject cloudy, colored, or particle-containing solution.
- With the separate injection syringe, the final daily volume is 0.16 mL. Retain the vial for seven consecutive daily doses.1
EGRIFTA SV: prepare each dose
2 mg vial + 0.5 mL supplied diluent- Use the supplied sterile water. Its bottle contains more liquid than one preparation needs.
- Measure 0.5 mL with the mixing equipment and transfer it into one vial.
- Discard the mixing syringe and needle. Roll the vial gently for 30 seconds, without shaking.
- Check that the liquid is clear, colorless, and particle-free.
- Prepare 0.35 mL using the new injection equipment specified in the kit; administer immediately. Discard leftover medicine and diluent.2
These preparation summaries accompany the complete illustrated instructions linked in the references. They are specific to the named medicines.
How long does mixed tesamorelin last?
| Product | After mixing | Discard point |
|---|---|---|
| EGRIFTA WR | 20–25°C, protected from light; do not refrigerate or freeze. | Seven days after mixing. |
| EGRIFTA SV | Use immediately; do not refrigerate or freeze. | Discard any unused prepared solution. |
Product instructions: WR1 and SV2.
Write the mixing date where it can be checked. A clear appearance does not prove that a solution remains sterile or potent beyond its stated period. Refrigeration is not a universal rule for every peptide formulation, and a generic “28 days after mixing” rule does not apply to these medicines.
Tesamorelin protocols and preparation FAQ
Do people use tesamorelin five days per week?
Yes. One cited commenter reported changing to 1 mg on five days each week after a higher IGF-1 result. Other accounts describe daily use. At the same amount per administration, five days produces a smaller weekly total; the reports do not establish that both schedules give equal results.C5
Is a 10 mg vial the daily dose?
Ten milligrams is the stated total in the container. A daily amount, concentration in mg/mL, and measured volume are separate quantities. The calculator converts an amount you enter; it does not select a regimen.
Is an eight-week update the same as an eight-week cycle?
Does a preloaded pen need reconstitution?
A pen already containing solution has no powder-mixing step. Its delivered volume per click must come from the device specification. The total milligrams alone do not reveal the click setting.
Can the same amount be used for a tesamorelin–ipamorelin blend?
A blend contains two separate amounts in a stated ratio. Community accounts should identify each one, as the reports above do. A combined milligram figure cannot be treated as the tesamorelin amount.
What if a blood test or side effect changes during a cycle?
Review the actual result and symptoms with a clinician before changing the regimen. The label calls for glucose assessment and IGF-1 monitoring. Persistent swelling, hand numbness or a troublesome injection reaction warrants assessment. See the side-effects guide for details.1
